Green carrot-callus cultures exposed for 3 h in liquid media to light and 14CO2 secreted a constant 68 per cent of their total 14C-photosynthate into the medium over a wide range of 14CO2-fixation rates, obtained by varying 14CO2 levels. The major secreted products (sucrose and glutamine) or analogues of these compounds did not affect secretion when included in the bathing medium. Dual-labelling studies with [6-T] glucose and 14CO2 demonstrated that secretion occurred from a metabolic rather than a storage compartment. The effects of long preincubation periods in liquid media and of osmotic shock showed that secretion took place from specific sites on the boundary of the metabolic compartment.
