The sucrose transporter, OsSUT1, is expressed in germinating rice embryos, where it is present in the scutellar vasculature and the vasculature of the coleoptile and primary leaves. An OsSUT1 promoter:GUS fusion was used to detect expression in whole seedings during a time-course through germination, a GUS-stained whole seedling at 5 d post-imbibition (dpi) is shown in the top panel. The middle panel shows a transverse section through the coleoptile, which contains the first and second leaves within it, viewed under darkfield illumination. GUS crystals, in red, are present in the vasculature of these tissues. The lower panel shows a transverse section through the same region of another seedling at 5 dpi that has been probed with an anti-OsSUT1 antibody. Antibody labelling of OsSUT1 epitopes in the coleoptile and primary leaves vasculaature is indicated by dark blue colouration. (See Scofield et al., pp. 483–495.)
